Abstract

The Sumbawa horse (Equus caballus) is one of the local Indonesian horse breeds spread across the province of West Nusa Tenggara which has uniform physical form, genetic composition, good physical ability in terms of environmental limitations and superior carrying capacity, drag and run. The development of horse livestock needs to be followed by efforts to conserve horse germplasm through an inventory of genetic resources to prevent inbreeding. The aims of this study were: (1) to describe the kinship relationship of the Sumbawa horse (Eqqus caballus) based on microsatellite DNA markers, (2) to determine the validity of the enrichment material for genetics courses. This is a descriptive exploratory researchusing 25 individual Sumbawa Horses as the samples. DNA isolation was carried out using KIT. Horse blood was taken through the jugular vein in the neck using a venoject and a vacuum (empty) tube. The tube that already contains blood is then given 0.1 gram of EDTA so that the blood does not clot. The results of the phylogenetic study of the Sumbawa horse are: (1) based on the results of cluster analysis it is known that individual horses in the West Sumbawa area are divided into several groups, namely group 1 KD11, KD13, KD20, KD21, KD23, KD12, KD14, KD16, KD17, KD18, KD19, KD22, KD24, and KD15, group 2 namely KD2, KD10, KD25, KD1, Group 3 namely KD3, KD5, KD7, KD8, KD9, KD4, and KD6. The grouping of individual horses into three clusters indicates a kinship relationship between the individual horses. This is presumably due to the occurrence of intermarriage among the horse population which is carried out by breeders through trade, resulting in genetic mixing between the horse individuals. Results Pocket book validation scoring based on the feasibility of content, language, and display respectively at 3.4; 3,4; 3,2 and all fall into the very good category.
