Abstract

The Sumbawa Horse (Equus caballus) is one part of the local Indonesian Horse family which is native to Sumbawa Island. The Sumbawa horse is a livestock genetic resource that must be preserved. Development of Sumbawa Horse livestock needs to be followed by efforts to conserve germplasm through identification of genetic variations. This needs to be done as a consideration in conducting outbreeding to obtain superior Sumbawa Horse seeds. The step that needs to be done is to analyze genetic diversity by looking at its kinship relationships. Information about the diversity/genetic diversity of the Sumbawa Horse needs to be used as enrichment material in the form of a pocket book. The purpose of this study was to describe the genetic diversity/diversity of the Sumbawa horse through the primary application of buffalo microsatellite and to determine the level of validity of the research results which can be used as enrichment material for genetics courses. This is a descriptive explorative research using 25 Sumbawa Horses as the samples. DNA isolation was carried out from a blood sample of the Sumbawa Horse taken from the jugular vein. This study used a primary type of buffalo microsatellite, namely HEL09. The results of DNA band analysis from PCR converted into binary data showed that microsatellite DNA alleles were found with lengths of 200 bp, 300 bp, 400 bp, 700 bp, 1000 bp and 1200 bp. Furthermore, the results of cluster analysis showed that there were 25 Sumbawa Horse individuals from individuals with the same population. These results indicate that in general buffalo microsatellite primers can reveal genetic diversity/diversity of the Sumbawa horse through DNA fingerprinting, but buffalo microsatellite primers cannot reveal alleles of the Sumbawa horse microsatellite. The validation results of the three validators, i.e the content, language and display validators, the level of validity of the research results in the form of a pocket book can be categorized as very good/feasible to be used as material enrichment for genetics course.
